چکیده

Background The human designer cell line AGE1.HN represents a promising production system for biopharmaceuticals, particularly for those needing human-type post-translational modifications [1,2]. For further rational improvement of the cell line and the cultivation process a detailed understanding of the metabolism and metabolic changes during glycoprotein production is desirable. Metabolism and metabolic burden upon production of a1-antitrypsin (A1AT) were analyzed by comparing parental cells and a derived clone, AGE1.HN.AAT. The questions addressed were (i), which changes occur in cell growth and metabolism upon A1AT production and (ii), what are specific cellular properties distinguishing the producer clone AGE1.HN.AAT from the parental cell population.

Quantitative characterization of metabolism and metabolic shifts during growth of the new human cell line AGE1.HN using time resolved metabolic flux analysis

For the improved production of vaccines and therapeutic proteins, a detailed understanding of the metabolic dynamics during batch or fed-batch production is requested. To study the new human cell line AGE1.HN, a flexible metabolic flux analysis method was developed that is considering dynamic changes in growth and metabolism during cultivation. This method comprises analysis of formation of cel...

Compartmentation and channelling of metabolites in the human cell line AGE1.HN®

Background A thorough knowledge of the metabolism and its compartmentation in mammalian cells is desirable to enable rational design and optimization of producing cell lines and production processes for biopharmaceuticals. In this study we focused on acquiring a detailed understanding of metabolite channelling and the metabolic flux distribution during overflow metabolism in the human cell line...

Physical methods for synchronization of a human production cell line

Background The study of central metabolism and the interaction of its dynamics during growth, product formation and cell division are key tasks to decode the complex metabolic network of mammalian cells. For this purpose, not only the quantitative determination of key cellular molecules is necessary, but also the variation of their expression rates in time, e.g. cell cycle dependent gene expres...

Correlation of α1-antitrypsin (A1AT), Complement component C5a and Secretory Immunoglobulin A (sIgA) with pulmonary complications; 20 years after sulfur mustard exposure, Sardasht-Iran Cohort Study

Background: Little is known about pulmonary complications induced by Sulfur Mustard (SM), as a chemical warfare agent, especially considering its long term effects. The present study was carried out to investigate the association of α1-antitrypsin(A1AT), Complement component C5a, and Secretory Immunoglobulin A (sIgA) with long-term pulmonary complications on SMexposedindividuals, 20 years after ...

Alpha- 1 Antitrypsin Deficiency in Children: Pulmonary Involvement

Introduction: α1-antitrypsin deficiency (α1-ATD) is one of the most common genetic disorders in white race, a usual cause of liver disease in children, and hepatopulmonary involvement in children and adult. The aim of this case description is presenting a child with early lung disease without liver parenchymal disorder. Case presentation: We describe a 13 year old boy because of exertional dysp...
